Formalin fixation and paraffin embedding (FFPE) is a method of choice for archiving human tissue for diagnostic purposes, whether in block or histological slide form, as it allows for long-term storage at ambient temperature while preserving structural and cellular integrity. This preservation method also enables subsequent analysis of the DNA and RNA contained within the fixed tissues, thus being valuable in forensic cases when no other genetic material from an individual remains. However, although several studies comparing the performance of various FFPE DNA extraction methods have been published, standardized methods for forensic DNA-based applications are lacking. The Laboratoire de sciences judiciaires et de médecine légale (LSJML) has therefore set out to provide a method to improve DNA typing results from FFPE samples originating from a variety of criminal cases by testing and validating a commercially available nucleic acid extraction kit (MagMAX TM). We present the results of three individual identification cases and one paternity case that required DNA extraction and STR typing of FFPE blocks and stained slide sections. This work highlights the value of FFPE samples in various criminal cases, the robustness of the method for extracting DNA and generating STR typing results, as well as the relevance of the improved method for other forensic laboratories seeking to validate and implement an FFPE extraction method.
