Polymerase chain reaction-massively parallel sequencing (PCR-MPS) has demonstrated advantages over standard PCR- capillary electrophoresis (PCR-CE) for forensics short tandem repeat (STR) profiling, as it can detect sequence-level variations that enhance discriminatory power between individuals. The Precision ID GlobalFiler™ NGS STR Panel v2 is a widely available commercial solutions for such forensic analyses. However, for laboratories to effectively use this technology, it is crucial to establish population-specific databases of sequenced allele frequencies, which are essential for accurate match reporting.
This study aimed to identify sequenced-based STR variant alleles within the Chinese population in Singapore, using Precision ID GlobalFiler™ NGS STR Panel v2 Kit on the Ion S5™ Sequencer. The objectives included generating sequence-based STR allele frequencies and comparing the diversity of sequence-based STR alleles to that of traditional length-based alleles. Sixty-four anonymised DNA samples were obtained from unrelated volunteers, who self-identified as Chinese in Singapore and provided informed consent. DNA was extracted from blood samples preserved on FTA cards. All samples underwent quantification, amplification using the Precision ID GlobalFiler™ NGS STR Panel v2, and sequence analysis using the accompanying CONVERGE™ software. This initial population-specific dataset offers foundational data to support the implementation and evaluation of sequenced-based STR profiling in Singapore’s forensic laboratories.
